RNAi inhibits, or suppresses, the expression of a specific gene of interest. In the RNAi pathway, short double stranded siRNA segments of about 20 base pairs act to block a cell’s RNA, in order to prevent the creation of destructive proteins caused by diseased or damaged DNA. The siRNA must be complementary to the target protein/gene.

The RNAi pathway has been proposed and studied for a wide range of therapeutic uses, including anti-viral therapy and treatments for cancer, liver disease, macular degeneration, and neurodegenerative disease, though off-target effects remain significant. RNAi may also be important in the genetic engineering of plants to reduce toxins or increase beneficial components, such as antioxidants.
